چکیده

Structural requirements for antifreeze and antilectin activities of the antifreeze glycoproteins from the Antarctic fish Trematomus borckgrevinki have been compared. Graded removal of different amounts of the disaccharide side chain by p elimination caused similar reductions in the capacities for depressing the freezing temperature of water (antifreeze activity) and for inhibiting hemagglutination by Osageorange lectin (antilectin activity). Both activities were also reduced to nearly similar extents by acetylation of hydroxyls of carbohydrate residues, positioning a negative charge on the C-6 carbons of the carbohydrate, or by formation of borate complexes. But oxidations of the C-6 hydroxyls of the carbohydrate to aldehyde groups caused no reductions of either activity. Structural similarities required for both activities therefore exist.

Antifreeze glycoproteins from Arctic fish.

Four species of fish in the Barents Sea of the Arctic, north of Norway and Russia, were caught and examined for the presence of antifreeze proteins in their blood sera. Only one species of Arctic fish, the polar cod (Boreogadus saida), was found to contain an antifreeze protein. Antifreeze glycoproteins were isolated and characterized from the blood serum of the Arctic fish, and their structure...

Antifreeze peptide heterogeneity in an antarctic eel pout includes an unusually large major variant comprised of two 7 kDa type III AFPs linked in tandem.

The structural heterogeneity of the major antifreeze peptides (AFPs) from the antarctic eel pout, Lycodichthys dearborni (formerly classified as Rhigophila dearborni) was characterized. Three major AFPs designated as RD1, RD2 and RD3, and five minor ones were isolated from the fish plasma. RD1 and RD2 are both 64 residues in length, about 7 kDa, and thus similar in size to all characterized typ...

Evolution of antifreeze glycoprotein gene from a trypsinogen gene in Antarctic notothenioid fish.

Freezing avoidance conferred by different types of antifreeze proteins in various polar and subpolar fishes represents a remarkable example of cold adaptation, but how these unique proteins arose is unknown. We have found that the antifreeze glycoproteins (AFGPs) of the predominant Antarctic fish taxon, the notothenioids, evolved from a pancreatic trypsinogen. We have determined the likely evol...

Purification and primary sequences of the major arginine-containing antifreeze glycopeptides from the fish Eleginus gracilis.

An Arg-containing antifreeze glycoprotein from the polar fish Eleginus gracilis was isolated, and the major components were purified to homogeneity. The general protocol for purification was chromatography of serum on DEAE-cellulose, followed by chromatography on a cation exchanger. DEAE-cellulose chromatography resulted in two fractions, A and B. Fraction A contained most of the antifreeze gly...
